That's the opening scene. Locke was *re-reading* the dossier, which he
had read for the first time two days prior.  Cooper is already locked
in the brig by then.  We never see how he got there, because the
illusion is that it's Ben in the brig.  After Locke burned the folder,
he would have set out to find Sawyer.

 [The scene opens with Locke sitting by a fire reading the contents of
a red folder. We later learn he is inside the Black Rock and that
someone is bound and gagged in the brig. They are screaming]
Locke: Save your breath. Nobody's gonna hear you.
  [He drops the folder into the fire which is burning in a metal can]

He had already read the dossier, two days before the killing took
place:

[2 Days Ago: The scene flashes back to Locke sitting on a hill staring
at the Others' tents. He unwraps his wounded hand that was bit by
Anthony and it is healed. Richard walks up]
Richard Alpert It's beautiful isn't it? No matter how much time you
spend on this island, you just never get tired of this view. We
haven't been formally introduced, I'm Richard.
  [He offers his hand to Locke who shakes it]
Richard Alpert You mind if I...join you here?
Locke Yeah, sure.
  [Richard sits on the ground by Locke]
Richard Alpert He wanted to embarrass you.
Locke I'm sorry.
Richard Alpert Ben knew you weren't going to kill your own father. He
put you in front of everyone in our camp just so they could all watch
you fail.
Locke Why?
Richard Alpert Cause when word got back here that there was a man with
a broken spine on the plane who could suddenly walk again, well,
people here began to get very excited because that...that could only
happen to someone who was extremely special. Ben doesn't want anyone
to think you're special, John.
Locke Why are you telling me this?
Richard Alpert Ben has been wasting our time with novelties like
fertility problems...we're looking for someone to remind us that we're
here for more im****tant reasons.
Locke What do you want from me?
Richard Alpert I want for you to find your purpose...and to do
that...your father has to go, John. And since you're not gonna do
it...
  [Richard reaches into a satchel on his shoulder and pulls out a red
folder which he hands to Locke]
Richard Alpert ...I'm gonna suggest someone else.
  [Locke opens the folder and reads as Richard gets up and walks
away]
Locke Sawyer? Why would Sawyer kill my father? He doesn't even know
him?
Richard Alpert Keep reading.

Yes, I realize that, but that's the scene he was describing.
Obviously he had no idea where that room was.
